Research summary
The objective of this research project is to establish an accurate methodology of measuring LDL/HDL fractions within total cholesterol from blood using a lateral flow system. We are utilizing an established method commonly used for quantification of LDL and HDL in solution and transitioning the protocol onto paper. The assay involves the precipitation of the LDL/VLDL fractions of total cholesterol. The resulting non-HDL fraction is then party to an enzymatic chemical reaction which allows it to be quantified. The LDL fraction in turn can then be calculated from the total cholesterol and HDL numbers. The reagents (precipitating agents, enzymes, stabilizers) concentrations and placement on the paper need to be established and optimized. In order to verify the effectiveness of the precipitation step and the overall assay, we need a number of clinical samples spanning the physiological range for LDL, HDL and total cholesterol.
