    Some children are affected by illnesses due to infection despite availability of antibiotics and vaccines. There is evidence that genetic factors can determine both susceptibility to and outcome after infection. The present study proposes that specific genetic defects may predispose individuals to suffer from rare, severe, or unusual presentations of infection. This study aims to investigate possible changes in one gene or a collection of genes that causes an increased risk for these conditions. Identification of rare genetic factors will improve our understanding of the body’s response to infection and may allow scientists to develop new diagnostic tests and treatments. It may also help doctors find new strategies to prevent infection and guide genetic counselling for families who are affected.
    Any patients who have a history of suffering from or who have an acute presentation with rare, severe, or unusual presentations of infection may be included. The focus will be on the severity of the illness and/or unusual appearance of infection. Other family members may also be invited to participate in this study (for example child’s biological parents, siblings, grandparents, aunts, uncles and cousins). We will also recruit control individuals with similar phenotypes and genotypes.
    The study will be conducted from Imperial College London. Sites where families are recruited will be NHS hospital sites from around the UK or from investigators from hospitals abroad. Whilst the study is open, participants (index cases) may undergo blood sampling on more than one occasion. In some cases a skin biopsy will be collected. Saliva, nails or hair may also be collected.
