    Oral lichen planus (OLP) is a chronic inflammatory disease of oral mucosa that is mediated by certain mediators of the immune system known as interleukins. Objectives: This research is a novel study as it is the first one to include Interleukin 27 and 28b/Interferon lambda 3 (IL28b/IFN ʎ3) and their role in the pathogenesis of (OLP) ,in addition to Interferon Gamma (IFN-γ) and Tumour Necrosis Factor-alpha (TNF-α) which were included in previous studies. The protein expression of the included interleukins will be studied and correlated with the clinical outcome as well as with their gene expression. Chronic inflammatory infiltrate cells will be isolated and analysis ex-vivo of IFN-γ and TNF-α production after stimulation with IL-27 or IL-28b/IFN ʎ3. Design: Tissue, blood and salivary samples will be taken from 50 patients with OLP (25 with reticular and 25 with erosive varieties) and 50 sex and age matched patients with other oral keratosis. The laboratory techniques to be used will include: immuno-histochemistry, quantitative real-time polymerase chain reaction PCR, sandwich enzyme linked immunosorbent assay ELISA, fluorometric detection as well as oral mucosa experimental model will be generated to study the immunopathology of OLP, with particular reference to IL-27 and IL-28b/IFN ʎ3 role and to study the GVHD reaction (graft versus host disease) that may resemble OLP clinically and histopathologically. The results will be correlated with a clinical database that contains details of the patient demographics, clinical presentation and outcome during treatment.
