Research summary
Aortic stenosis (AS) occurs when one of the heart's valves narrows and blood cannot flow to the rest of the body normally. The condition can range from mild to severe and AS progression is unpredictable often occurring over many years. Current treatments are limited to replacement of the faulty valve. Timely replacement of the faulty valve before the level of heart damage becomes too severe is essential however, predicting which patients will benefit from valve replacement and when to intervene is currently challenging due to lack of reliable clinical tools. Genetic material circulating in our blood can provide important information related to disease severity. This information is easily accessible and can be found freely circulating in the blood or specifically associated with tiny particles, known as extracellular vesicles (EVs). EVs differ in size and function and can be separated into small and large EVs with both having important biological functions. In this project, we will follow patients before and after valve replacement to identify which patients respond well to valve replacement according to their heart function and symptoms. We will analyse the genetic material circulating in patient blood and specifically associated with small and large EVs to identify new predictors of patient outcome. Together this will be an important step towards the development of a much needed tool for AS disease management that can be used to identify the highest risk individuals for closer monitoring and timely intervention.
