    In this research study we want to learn more about how genes and the environment affect how young people recover from traumatic brain injury (TBI). Precision medicine is an approach for disease treatment that takes into account individual variability in genes, environment, and lifestyle for each person. There is significant variation in recovery from paediatric TBI and we lack understanding of how genetic and home and family environmental factors may influence this. If we could understand what causes this variation, then treatments could be optimised to improve how people recover from these injuries. \n\nThis project will examine the associations of genes and environment with outcomes after severe paediatric TBI. We predict that genetic variants that are important to biological processes involved in TBI recovery will interact with environmental factors to influence recovery after severe paediatric TBI. Findings will help to understand individual prognoses for young people with TBI and develop novel, precision treatments to improve their recovery.\n\nWe will invite children and families in England who are already participating in an existing international paediatric TBI trial, the Approaches and Decisions in Acute Paediatric TBI Trial, to take part in this study. We will ask young people to provide a saliva sample to collect genetic information and ask their parent/guardian to complete short questionnaires about their home and family environment. Families can do this at home in approximately 30 minutes and return the saliva sample and questionnaires using a freepost envelope provided by the research team. Telephone support will be available from the research team. Participation will end when the saliva sample and questionnaire are returned. The study will end in April 2021 and participating families will receive a newsletter informing them of the research findings.\n\nThis research is funded by the National Institutes of Health (NIH).\n
