    We have planned a prospective study of patients with CIS who will be studied with MRI, OCT, and clinical assessments at study entry and after 6 and 12 months. Patients will have their bloods collected at baseline for a genetic analysis and a sub-group of patients will be recruited to a PET pilot study. As well as providing substantial insights into early pathological changes both at the lesion site and within the brain in demyelinating CIS, this investigation will determine which early changes in imaging and OCT metrics can predict later clinical conversion or disease activity at a more subject-specific level. PET imaging will help to eludicate in-vivo the role of the innate immune response using PET scanning, to see if the activation of microglial cells is necessary for the conversion to MS in patients who present with a clinically isolated syndrome. This will help to inform prognosis and target future therapies in CIS.
